Study on the medication rule and mechanism of core prescription of Chinese herbal compound in the treatment of irritable bowel syndrome based on patent database

Abstract:
Objective To analyze the medication rule and action mechanism of compound Chinese medicine(TCM) patent prescriptions for irritable bowel syndrome, and to provide a reference for new drug development and patent protection. Methods The patent announcement of State Intellectual Property Office(http://epub.sipo.gov.cn) was searched by computer from September 1985 to July 2022, the data of TCM patent prescriptions for the treatment of irritable bowel syndrome were imported into the ancient and modern medical records cloud platform V2.2.3 for medication frequency analysis, association analysis and cluster analysis, and core prescriptions were extracted through complex network analysis. The active ingredients and targets of core prescription drugs were screened and standardized by TCMSP, Batman-TCM, SwissTargetPrediction, Uniprot and other databases, and the composition-target network diagram was constructed by Cytoscape. The disease targets were obtained from GeneCards database platform, and common targets of drugs and diseases were obtained. Finally, GO enrichment analysis and KEGG signaling pathway analysis were performed on the common targets of drugs and diseases using R language. Results 72 TCM patent prescriptions were included, involving drugs 317, the most commonly used drugs for Chenpi(陈皮), Baishao(白芍), Baishu(白术), Gancao(甘草), Fangfeng(防风), etc. Commonly used drugs category mainly have the functions of heat-clearing and detoxifying, drying dampness and resolving phlegm, regulating qi to relieve pain, regulating qi-flowing for streng-thening spleen, regulating liver to relieve pain, etc. Drugs mainly occur among the herbs that are warm or mild in property, sweet, pungent or bitter in taste. Herbs mainly distribute to Pi(脾), Wei(胃) and Fei(肺) jing. The results of Assoc-iation analysis showed that the most commonly used medicine suits were Baishu-Baishao(白术-白芍), Fangfeng-Chenpi(防风-陈皮). The results of cluster analysis divided the included high-frequency drugs into 6 categories. Finally, 131 kinds of drug components were screened out, 263 potential targets were predicted, and 52 common targets of core prescriptions and diseases were mainly enriched in 1 816 biological processes and 146 signaling pathways. Conclusion The medication rule of TCM patent prescriptions in treating irritable bowel syndrome is consistent with clinical practice and its mechanism of action is consistent with pathological mechanism of irritable bowel syndrome.
