Effects of Bazhen Decoction on the Expression of EGF, TGF-β1 and VEGFA in Cultured Human Skin Fibroblasts

Abstract:
Objective Observe the effect of Bazhen Decoction of different concentrations on the expression of EGF, TGF-β1 and VEGFA in cultured human skin fibroblasts in vitro. Methods The CCK-8 method was used to determine the effect of different concentrations of Bazhen Decoction on the proliferation of human skin fibroblasts.The drug concentration and medication time were determined. Western blotting was used to detect the effects of different concentrations of Bazhen Decoction on the expression of EGF, TGF-β1 and VEGFA in human skin fibroblasts. Results Bazhen Decoction can promote the proliferation of human skin fibroblasts, and the effect of promoting cell proliferation is obvious when the concentration is 1 mg/L-1 g/L. The effect of drug intervention in promoting cell proliferation is obvious for 24 hours (P<0.01). Compared with the control group, the expressions of EGF, TGF-β1 and VEGFA in the medium concentration Bazhen decoction (100 mg/L) group were significantly increased (P<0.05). The expression of EGF and TGF-β1 in low concentration Bazhen Decoction (10 mg/L) group were not significantly increased (P>0.05). The expression of VEGFA was increased (P<0.05). The expression of EGF and TGF-β1 in the high-concentration Bazhen Decoction (1g/L) group decreased (P<0.05), and the expression of VEGFA was not significant (P>0.05). Conclusion The medium concentration Bazhen Decoction can accelerate the proliferation of human skin fibroblasts, promote the expression of EGF, TGF-β1 and VEGFA, thereby accelerating angiogenesis and promoting the growth of wound granulation tissue, and ultimately promote wound healing.
