Differential Study on Dynamic Changes of Inflammatory Factors in Diabetic Rats and Normal Rats at Different Stages of Wound Healing

Abstract:
Objective To observe the differences in dynamic changes of wound inflammation-related factors in diabetic rats and normal rats at different stages during wound healing, and to explore the influence factors of the healing of diabetic ulcers. Methods By intraperitoneal injection of Streptozocin and surgical method to establish the rat model of diabetes ulcers. The different expressions of various detection indexes were observed and compared including macrophages, interleukins, TNF-αof diabetic model and normal control groupby the methods of immunohistochemistry. Results The number of M1-type macrophages of diabetic ulcer model group was significantly lower in the inflammatory stage, while the number of M2-type macrophages was significantly lower in the regenerated stage. It is different from the normal control group that the levels of IL-1 of the diabetic ulcer model group inthe regenerated stage[(7.169±0.785)%]were significantly higher than those in the inflammatory stage[(6.038±0.939)%] (P<0.05).The levels of TNF-αof the diabetic ulcer model group in the regenerated stage[(8.651±0.646)%]were significantly higher than those in the inflammatory stage[(4.860±0.654)%] (P<0.01). The levels of IL-6 of the diabetic ulcer model group inthe regenerated stage[(9.627±1.211)%]were significantly higher than those in the inflammatory stage[(5.457±0.643)%] (P < 0 . 01 ) while the ascensional range of IL-10 were significantly smaller than those of the normal group. Conclusion The reasons of delayed healingof diabetic ulcer may be related to abnormal phenotype and transformation of macrophage, and the dynamic imbalance between proin?ammatory factors and anti-in?ammatory factors.
