Effect of Taohe Chengqi Decoction on LPS-induced Expression of Vascular Endothelial Inflammatory Factors, TLR4 and TRIB3

Abstract:
Objective To observe the effect of Taohe Chengqi Decoction on the expression of inflammatory factors, TLR4 and TRIB3 secreted by vascular endothelium in vitro induced by LPS. Methods Preparation of drug-containing serum by gavage of SD rats with Taohe Chengqi Decoction. LPS induced aortic vascular endothelial cell injury model. They were divided into normal group, normal dosing group, model group and model dosing group. The expression levels of IL-1β, IL-6 and TGF-β in each group were compared by ELISA. The relative mRNA expression levels of IL-1β, IL-6, TGF-β, TLR4 and TRIB3 in each group were measured by qPCR. Results The expressions of IL-1β, IL-6 and TGF-β were increased in the model group compared with the normal group. Compared with the model group, IL-1β and IL-6 proteins in the other three groups were significantly inhibited, and TGF-βprotein expression in the model group was significantly increased, which the differences were statistically significant (P<0.05). Compared with the model group, the expression of IL-1β mRNA in the model group was inhibited, the expression of IL-6 mRNA and TLR4 mRNA in the other three groups were decreased, while the expression of TGF-β mRNA in the normal group and the normal group were increased, which the differences were statistically significant (P<0.05). Conclusion Taohe Chengqi Decoction contains serum that can inhibit the expression of IL-1β and IL-6, promote the expression of TGF-β and it plays a protective role on vascular endothelial cells, in which TLR4 signaling pathway may be involved.
