Study on Therapeutic Mechanism of Epimedium on Ulcerative Colitis in Rats

Abstract:
Objective To observe the therapeutic effect of epimedium on ulcerative colitis in rats and the effect on the expression of related inflammatory factors in colon tissue. Methods 84 male SD rats were randomly divided into blank control group, model group, epimedium high, medium and low dose groups, sulfasalazine (SASP) group and combination treatment group, with 12 rats in each group. Disease activity index (DAI), gross colon injury score, colon histopathology score, colon tissue TNF-α, IFN-γ and IL-6 immunohistochemical expression were used to evaluate its therapeutic effect. Results On the 4th day of the experimental treatment, the rats in each experimental group gained weight compared with the model group, and the rats in the combination group gained weight compared with the SASP group on the 7th day of the experiment,which the differenceswere statistically significant (P <0.05). From the beginning of the day, the DAI scores of each experimental group increased compared with the model group, which the difference was statistically significant (P <0.05). The degree of colon injury in each experimental group was compared with that in the model group,the colon ulcer area and the ratio of colon weight to length in the combined group decreased. Epimedium high,medium, and low dose groups and the combination group had less colonic damage. The area of colon ulcers in the combination group was smaller than that of the SASP group, which the difference was statistically significant (P <0.05). Compared with the model group, the epimedium colonic histopathological score of Huo in high-dose group was significantly reduced (P <0.01).The expression levels of TNF-α, IFN-γ and IL-6 in the colon tissue of rats in each experimental group were significantly reduced in the combination group and SASP group compared with the model group (P <0.01), and the combination group was significantly lower than those of the SASP group (P <0.01). Conclusion Epimedium can improve the mental state of rats with ulcerative colitis and reduce the degree of colon damage. Its therapeutic effect may be related to the reduction of TNF-α, IFN-γ and IL-6 cytokine expression.
