Effect and Mechanism of Taraxasterolon Migration and Invasion of T24 Bladder Cancer Cells CHEN

Abstract:
Objective To explore the effect and mechanism of taraxasterol on T24 bladder cancercell migration and invasion. Methods T24 bladder cancer cells were cultured in vitro and then differentconcentrations (10, 20, 40 μg/mL) of taraxasterol were added. The migration and invasion abilities of bladder cancer cells were observed by the scratch test and the transwell assay. Western blotting assay was performed to investigate the changes of SDF-1/CXCR4 signaling related protein expressions. The level of MMP-2/9 in cell supernatant was detected by ELISA method. Results Taraxasterol inhibited the migration and invasion of T24 bladder cancer cells (P < 0.05, P < 0.01) and decreased the ability of cells to secrete MMP-2 and MMP-9 (P < 0.05, P < 0.01). Meanwhile, taraxasterolcan effectively inhibit the expression levels of SDF-1, CXCR4, p-Akt and p-mTOR in T24 cells (P < 0.05, P < 0.01). Conclusion Taraxasterolcould interfere with SDF-1 /CXCR4 signal, down-regulate the Akt/mTOR conduction and weaken the ability of T24 cells to secrete MMP-2 and MMP-9. Finally, taraxasteroleffectively inhibit the migration and invasion of T24 bladder cancer cells.
