舒筋定痛胶囊通过调控NOD样受体家族含pyrin结构域蛋白3炎性小体治疗快速老化P6品系小鼠骨质疏松

摘要:
目的:研究舒筋定痛胶囊治疗快速老化P6品系(SAMP6)小鼠骨质疏松的疗效及其机制。方法:16只SAMP6小鼠随机分为空白对照组(SAMP空白组)和舒筋定痛治疗组(SJDT治疗组),每组各8只,8只抗快速老化小鼠(SAMR1)为SAMR对照组。SJDT治疗组每日灌胃舒筋定痛胶囊混悬液,SAMP空白组和SAMR对照组灌胃等量的生理盐水,8周后,采取双能X线吸收法(DXA)检测骨密度,微米X-射线显微镜(Micro-CT)检测骨微观结构;采用ELISA法检测血清中骨钙素(OCN)、I型胶原蛋白N端前肽(PINP)、抗酒石酸酸性磷酸酶(TRAP-5b)、I型胶原蛋白C端肽(CTX-Ⅰ)水平;采用Western blot和RT-PCR检测骨组织中NOD样受体家族含pyrin结构域蛋白3(NLRP3)、半胱氨酸天冬氨酸蛋白酶-1(Caspase-1)、Gasdermin D蛋白(GSDMD)、白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)蛋白和mRNA表达水平;采用免疫组化检测NLRP3表达。结果:与SAMP空白组和SAMR对照组相比,SJDT治疗组的骨矿物质密度(BMD)、骨体积分数(BV/TV)、骨小梁的厚度(Tb.Th)和骨小梁的数量(Tb.N)升高,骨小梁分离度(Tb.Sp)降低,差异有统计学意义(P <0.05);SJDT治疗组OCN、PINP表达水平升高,而TRAP-5b、CTX-Ⅰ表达水平降低,差异有统计学意义(P <0.05);SJDT治疗组中NLRP3、Caspase-1、GSDMD、IL-1β、IL-18蛋白和mRNA表达降低,差异有统计学意义(P <0.05)。结论:舒筋定痛胶囊能够通过抑制NLRP3炎性小体表达水平,减轻炎症反应,抑制破骨细胞活性,促进成骨细胞形成,提示NLRP3可能是治疗骨质疏松的潜在靶点。

Shujin Dingtong capsules treat osteoporosis in SAMP6 mice by regulating NLRP3 inflammasome

Abstract:
Objective To investigate the therapeutic effect and mechanism of Shujindingtong capsules on osteoporosis in SAMP6 mice. Methods Sixteen SAMP6 mice were randomly divided into a blank control group (SAMP blank group) and a Shujindingtong capsules treatment group (SJDT treatment group), and eight SAMR1 mice were used as the control group (SAMR control group). The SJDT treatment group was administered Shujin Dingtong capsules suspension by gavage daily, while the SAMP and SAMR groups were given the same volume of normal saline. After 8 weeks, bone mineral density (BMD) was measured by dual-energy X-ray absorptiometry (DXA), and bone microstructure was examined by micro-computed tomography (Micro-CT). The expression levels of OCN, PINP, TRAP-5b, and CTX-I in serum were detected by enzyme-linked immunosorbent assay (ELISA). The expression levels of NLRP3, Caspase-1, GSDMD, IL-1β, and IL-18 proteins and mRNAs in bone tissue were detected by Western blot and RT-PCR. The expression of NLRP3 was detected by immunohistochemistry. Results Compared with the SAMP blank group and the SAMR control group, the SJDT treatment group showed significant increases in bone tissue BMD, BV/TV, Tb.Th, Tb.N, and a significant decrease in Tb.Sp (P <0.05). The expression levels of OCN and PINP in the SJDT treatment group were increased, while the expression levels of TRAP-5b and CTX-I were decreased (P <0.05). The expression levels of NLRP3, Caspase-1, GSDMD, IL-1β, and IL-18 proteins and mRNAs in the SJDT treatment group were significantly decreased (P <0.05). Conclusion Shujin Dingtong capsules can inhibit the expression level of NLRP3 inflammasome, reduce inflammatory response, suppress osteoclast activity, and promote osteoblast formation, NLRP3 may be a potential target for osteoporosis therapy.
