洋金花对膝关节手术后关节腔内炎症反应的影响

摘要:
目的:挖掘GEO数据库中膝关节内免疫细胞数据,将洋金花组分与巨噬细胞相关差异基因(DEMRGs)构建组分-靶点网络图及分子对接,以探究洋金花组分能否有效抑制膝关节手术后关节腔内炎症反应。方法:从GEO数据库下载GSE279915数据集以及洋金花各组分靶点的相关数据。利用limma数据包和相关性分析,获得巨噬细胞相关差异表达基因,并进行通路富集分析、洋金花组分-靶点互作网络的构建。采用分子对接验证洋金花组分和获得的差异基因之间的关系。结果:基于膝关节单核/巨噬细胞的转录组分析,共鉴定出1538个具有统计学意义的差异表达基因(DEGs),其中679个基因显著上调,859个基因显著下调。使用韦恩图筛选得到洋金花作用相关靶点及DEMRGs的交集基因。进一步使用STRING11.0数据库对洋金花的DEMRGs靶点进行蛋白互作网络图的构建,前10个关键基因为TNF、IL-6、AKT1、ESR1、PTGS2、INS、IL-1Β、BCL2、CASP3、GSK3B。为验证生物信息学预测的可靠性,使用分子对接验证洋金花组分-靶点结合情况,结果发现洋金花组分芦丁、槲皮素均可以和白细胞介素(IL)-1β、IL-6以及肿瘤坏死因子(TNF)结合,从而发挥抗炎效果。结论:洋金花及其活性成分通过多靶点、多途径发挥抗炎和镇痛作用,在膝关节手术围手术期管理中具有潜在应用价值。

Abstract:
Objective To conduct data mining on immune cell data in knee joint in GEO database, and construct a component-target network map and molecular docking between components and macrophage associated differentially expressed genes (DEMRGs). Methods Downloading the GSE279915 dataset and related data on the components and their targets. limma data package and correlation analysis were used to obtain DEMRGs, and the pathway enrichment analysis and the composition-target interaction network were carried out to obtain the key genes. To detect the relationship between the components and the selected differential genes, we used molecular docking to verify the binding. Results Compared the transcriptomic data of monocytes and macrophages in knee joint, 679 up-regulated and 859 down-regulated differentially expressed genes were obtained. The intersection genes of associated targets and DEMRGs were obtained by using Venn diagram. The composition-target interaction network was constructed using STRING database. The top 10 Degree key genes were TNF, IL-6, AKT1, ESR1, PTGS2, INS, IL-1β, BCL2, CASP3 and GSK3B. Molecular docking results showed that Rutin and quercetin could bind to IL-1β, IL-6 and TNF, and thus exert anti-inflammatory effects. ConclusionYangjinhua and its active ingredients exert anti-inflammatory and analgesic effects through multiple targets and pathways, and have potential application value in perioperative management of knee joint surgery.
