Effect of Xiezhuo Jiedu Recipe on serum inflammatory cytokines and HIF-1α protein expression in colonic tissue of rats with ulcerative colitis

Abstract:
Objective To observe the effects of Xiezhuo Jiedu Recipe on the expression of inflammatory cytokines and hypoxia inducible factor 1α(HIF-1α) protein of ulcerative colitis (UC) rats. Methods TNBS/ethanol combined enema was used to establish UC rat models, which were divided into model group, mesalazine group, Xiezhuo Jiedu Recipe high-dose group, medium-dose group and model group were given normal saline intragastric administration for 14 days. After administration, the rats were killed, and the contents of interleukin(IL)-6, IL-10 and IL-17 in serum of the rats were detected by ELISA. The expression of HIF-1α protein in colon tissue was observed by Western blot. Results Xiezhuo Jiedu Recipe can improve the inflammatory response of the colonic mucosa in UC rats, compared with the model group, the pathological damage score of the rat colon mucosa in each dose group of the Xiezhuo Jiedu Recipe was significantly lower, the content of IL-10 in serum was significantly higher, the content of IL-6 and IL-17 was significantly lower(P <0.05).The expression of HIF-1α protein of UC rats could be decreased in each dose group of Xiezhuo Jiedu Recipe compared with model group (P <0.05). Conclusion Xiezhuo Jiedu Recipe can improve the pathological injury of colon mucosa in UC rats, and its mechanism may be related to regulating serum inflammatory cytokines and decreasing the expression of HIF-1α protein in colon tissue.
