桃核承气汤对热秘型功能性便秘大鼠肠道菌群的影响

摘要:
目的:探究桃核承气汤对热秘型便秘大鼠的肠道功能和菌群组成及丰度的影响。方法:将40只SPF级雌性Wistar大鼠随机分为5组:对照组、便秘模型组、低剂量桃核承气汤干预组、高剂量桃核承气汤干预组、乳果糖阳性干预组,每组8只。适应性喂养1周后,对照组不给予药物干预,其他四组大鼠均使用热性中药结合肠蠕动抑制剂灌胃给药14 d诱导建立便秘模型,其中使用低剂量(10 g/kg)和高剂量桃核承气汤(20 g/kg)进行干预治疗,另外一组进行乳果糖干预作为阳性对照。28 d灌胃结束后,将大鼠放置代谢笼24 h,收集大鼠新鲜粪便,取肠道组织后采集相关数据进行统计分析,用16S rRNA高通量测序检测大鼠粪便中的微生物后行定量分析。结果:桃核承气汤干预后,便秘大鼠的粪便颗粒数、含水率及肠道炭末推进率均显著升高。16S rRNA测序结果显示,Verrucomicrobiae、Actinobacteria、Akkermansiaceae等有益菌群相对丰度升高。代谢组学研究发现组间代谢物差异明显(P <0.05),对相关统计代谢物进行KEGG富集分析,结果显示筛选出的差异代谢物主要涉及6条代谢途径。结论:桃核承气汤可显著改善热性中药诱导的便秘模型大鼠的肠道功能,促进肠道微环境向健康状态转变,其作用机制可能与调节肠道菌群的结构及功能平衡有关。

Effects of Taohe Chengqi decoction on gut microbiota of rats with functional constipation

Abstract:
Objective To investigate the effects of Taohe Chengqi decoction on the intestinal function, composition and abundance of the intestinal flora in rats with heat-type constipation. Methods Forty SPF-grade female Wistar rats were randomly divided into 5 groups, with 8 rats in each group. The groups were set as the control group, the constipation model group, the constipation model + low-dose Taohe Chengqi decoction intervention group, the constipation model + high-dose Taohe Chengqi decoction intervention group, and the constipation model + lactulose positive intervention group. After one week of adaptive feeding, the control group was not given any drug intervention. In the other four groups, the rats were intragastrically administered with heat-natured traditional Chinese medicines combined with intestinal motility inhibitors for 14 days to induce the establishment of a constipation model. Among them, two groups were randomly set as experimental groups and were intervened and treated with low-dose(10 g/kg) and high-dose(20 g/kg) Taohe Chengqi decoction respectively, and another group was intervened with lactulose as a positive control. After the intragastric administration was completed for 28 days, the rats were placed in a metabolic cage for 24 hours, and fresh feces of the rats were collected. After taking tissues, relevant data were collected for statistical analysis. 16S rRNA high-throughput sequencing was used to detect the microorganisms in the feces of the rats, and quantitative analysis was carried out. Results After the intervention of Taohe Chengqi decoction, the number of fecal pellets, water content and intestinal charcoal powder propulsion rate of the constipated model rats increased significantly. The results of 16S rRNA sequencing showed that the relative abundances of beneficial flora such as Verrucomicrobiae, Actinobacteria, and Akkermansiaceae increased. Metabolomics studies found that there were obvious differences in metabolites between groups (P <0.05). KEGG enrichment analysis was carried out on the relevant statistical metabolites. The results showed that the screened differential metabolites were mainly involved in six metabolic pathways. Conclusion Taohe Chengqi decoction can significantly improve the intestinal function of constipated model rats induced by heat-natured traditional Chinese medicines and promote the transformation of the intestinal microenvironment towards a healthy state. Its mechanism of action may be related to regulating the structural and functional balance of the intestinal flora.
