Effect and mechanism of Qibai Yuyang oil on diabetic rat ulcer wound

Abstract:
Objective To observe the therapeutic effect of Qibai Yuyang oil on diabetic ulcer wound by using diabetic rat ulcer model, and to study the relevant mechanism. Methods Eighty male SD rats were randomly divided into a blank control group (n=16) and a diabetes group (n=64),The latter was used to prepare ulcer model of diabetes rats, After modeling, the mice were randomly divided into model group, Qibai Yuyang oil group, and compound comfrey oil group. The rats were randomly divided into blank control group, model group, Qibai Yuyang oil group and compound comfrey oil group. The changes of wound size were recorded during the experiment. HE staining was used to observe the morphological changes of ulcer wound tissue. The expression of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were detected by immunohistochemical staining. Results The wound healing rate of Qibai Yuyang oil group and compound comfrey oil group was significantly higher than that of model group (P<0.05). The wound healing rate of Qibai Yuyang oil group was higher than that of compound comfrey oil group (P<0.05). The expressions of IL-6 and TNF-α in wound tissue of Qibai Yuyang oil group and compound comfrey oil group were significantly lower than those of model group (P<0.05). At 7 days after drug intervention, the expression of IL-6 in the wound tissue of Qibai Yuyang oil group was significantly lower than that of compound comfrey oil group (P<0.05), but there were no significant differences in the rest. Conclusion Qibai Yuyang oil can reduce the inflammatory reaction of wound, reduce the overexpression of pro-inflammatory mediators IL-6 and TNF-α in wound tissue, and promote the healing of ulcer wound.
