Effect of ginsenoside Rg3 on PAS domain protein 2 on biological behavior of cervical cancer cells

Abstract:
Objective To explore the mechanism of PAS domain protein 2 (NPAS2) regulated by ginsenoside Rg3 on the biological behavior of cervical cancer cell. Methods Cervical cancer cell Hela was cultured regularly, and the interaction between LncRNA XIST and NPAS2 was detected by RNA pull-down and RIP. Cervical cancer cells Hela were then treated with ginsenoside Rg3 (0, 25, 50, 100 and 200 μmol/L) at different concentrations. Cell proliferation rate was detected by MTT assay, cell invasion was detected by Transwell, cell apoptosis was detected by flow cytometry, and the relative expression of LncRNA XIST was detected by RT-PCR. The relative expression of NPAS2 was detected by RT-PCR and Western blot. Results LncRNA XIST was found to interact with NPAS2 in Hela cells. Compared with the blank control group, the proliferation rate and invasion of ginsenoside Rg3 groups in 25 μmol/L, 50 μmol/L, 100 μmol/L and 200 μmol/L groups were significantly decreased, the apoptosis level was significantly increased (P<0.05). The expression of LncRNA XIST was decreased. The mRNA and protein expressions of NPAS2 were increased in a dose-dependent manner (P<0.05). Conclusion Ginsenoside Rg3 can decrease the expression of LncRNA XIST, promote the expression of NPAS2, and then affect the biological behavior of cervical cancer cell proliferation, invasion and apoptosis.
