杜仲调节JAK2/STAT3信号通路对去卵巢骨质疏松大鼠的作用及机制研究

摘要:
目的 研究杜仲对去卵巢骨质疏松症(OVX)大鼠骨的保护作用及机制。方法:将40只SD大鼠随机分为Sham组、OVX组、OVX+杜仲组、OVX+雌激素组,每组10只。Sham组大鼠仅切除卵巢旁部分脂肪组织,其余3组大鼠均切除双侧卵巢建立OVX模型。OVX+杜仲组给予杜仲颗粒剂2.76 g/kg灌胃,OVX+雌激素组采用炔雌醇10 μg/kg灌胃,Sham组及OVX组灌胃等量蒸馏水。显微计算机断层扫描(Micro CT)检测各组大鼠股骨小梁的微观结构[骨矿物质密度(BMD)、骨矿物质含量(BMC)、骨小梁体积分数(BV/TV)、骨小梁厚度(Tb.Th)、骨表面体积比(BS/BV)和骨小梁分离度(Tb.Sp)],小动物骨密度仪和骨强度检测仪检测股骨骨质情况,Western blot检测JAK2/STAT3通路蛋白含量,酶联免疫吸附测定(ELISA)法检测血清骨标志物骨碱性磷酸酶(BALP)、前胶原I型肽(PINP)、骨钙蛋白(OCN)、胶原酶降解肽(CTX-1)含量。结果:OVX大鼠BMD、BMC、BV/TV和Tb.Th明显降低,BS/BV和Tb.Sp明显升高(P <0.01),杜仲可显著提高BMD、BMC、BV/TV和Tb.Th,明显降低BS/BV和Tb.Sp(P <0.01),同时增强骨密度和骨压碎力。同时,杜仲下调OVX大鼠血清BALP和CTX-1水平,上调PINP和OCN水平(P<0.01)。机制探究显示,杜仲可调节OVX大鼠股骨中的JAK2/STAT3信号通路,降低p-JAK2和p-STAT3蛋白的相对表达水平(P <0.01)。结论:杜仲可增强OVX大鼠的股骨骨质,减轻骨质疏松严重程度,其机制可能与调节JAK2/STAT3通路有关。

Effect and mechanism of Eucommia ulmoides in regulating the JAK2/STAT3 Signaling pathway in ovariectomized osteoporotic rats

Abstract:
Objective:?To investigate the protective effects and mechanisms of Eucommia ulmoides on bone in ovariectomized(OVX) rats. Methods:?Forty SD rats were randomly divided into four groups: sham group, OVX group, OVX+Eucommia group, and OVX+estrogen group, with ten rats in each group. The sham group underwent partial removal of the fat tissue adjacent to the ovaries, while the other three groups underwent bilateral ovariectomy to establish the OVX model. The OVX+Eucommia group received a gavage of Eucommia granules at a dosage of 2.76 g/kg, and the OVX+estrogen group was administered ethynyl estradiol at 10 μg/kg via gavage. The sham and OVX groups were given an equivalent volume of distilled water. Micro-computed tomography (Micro CT) was used to assess the microstructural parameters of the femoral trabecular bone in each group, including bone mineral density (BMD), bone mineral content (BMC), trabecular volume fraction (BV/TV), trabecular thickness (Tb.Th), bone surface-to-volume ratio (BS/BV), and trabecular separation (Tb.Sp). A small animal densitometer and bone strength testing apparatus were used to evaluate the bone properties of the femur. Western blot analysis was conducted to measure the levels of JAK2/STAT3 pathway proteins, and enzyme-linked immunosorbent assay (ELISA) was performed to determine the serum levels of bone markers: bone alkaline phosphatase(BALP),procollagen type I N-terminal propeptide (PINP), osteocalcin (OCN), and collagen type I C-terminal cross-linked telopeptide (CTX-1).Results: The OVX rats exhibited significant reductions in BMD, BMC, BV/TV, and Tb.Th, alongside significant increases in BS/BV and Tb.Sp (P<0.01). Eucommia ulmoides significantly improved BMD, BMC, BV/TV, and Tb.Th, and markedly reduced BS/BV and Tb.Sp (P<0.01), while also enhancing bone density and strength. Furthermore, Eucommia ulmoides downregulated serum levels of BALP and CTX-1, while upregulating levels of PINP and OCN in OVX rats (P<0.01). Additionally, mechanistic investigations revealed that Eucommia ulmoides modulated the JAK2/STAT3 signaling pathway in the femur of OVX rats, leading to decreased relative expression levels of p-JAK2 and p-STAT3 (P<0.01). Conclusion:?Eucommia ulmoides can increase femoral bone quality and reduce the severity of osteoporosis in OVX rats, possibly through the modulation of the JAK2/STAT3 signaling pathway
