Study of prognostic predictive role of miRNA-28-5p in cholangiocarcinoma patients and its promotion of tumor proliferation and invasion

Abstract:
Objective To investigate the relationship between miR-28-5p expression and the prognosis of cholangiocarcinoma patients, as well as its functional role in tumor occurrence and development. Methods A total of 112 cholangiocarcinoma patients who underwent surgical resection were included in this study. The expression levels of miR-28-5p were detected in 112 pairs of cholangiocarcinoma tissues and adjacent non-cancerous tissues, as well as in CCA cell lines (TFK-1, RBE, HuH28, HuCCT-1, QBC939) and normal biliary epithelial cells (HIBEpic). The relationship between miR-28-5p and patient prognosis was analyzed using Kaplan-Meier curves and Cox regression analysis. The effects of miR-28-5p on cell proliferation, migration, and invasion were assessed using CCK-8 and Transwell assays. Results The expression levels of miR-28-5p in cholangiocarcinoma tissues and cell lines were significantly higher than those in adjacent non-cancerous tissues and normal epithelial cells. Additionally, low expression of miR-28-5p was significantly associated with poor prognosis in patients. Downregulation of miR-28-5p promoted proliferation, migration, and invasion of cholangiocarcinoma cells, while overexpression of miR-28-5p inhibited these processes. Conclusion miR-28-5p holds promise as a prognostic marker and potential therapeutic target for cholangiocarcinoma patients.
