miRNA-28-5p对胆管细胞癌患者预后的预测作用及对肿瘤增殖和侵袭的作用

摘要:
目的:探讨 miR-28-5p表达与胆管细胞癌患者预后的关系及其对肿瘤增殖和侵袭的作用。方法:纳入 2017年 10月— 2020年 12月在我院接受根治性切除的胆管细胞癌患者 112例。选取其胆管细胞癌组织和相邻癌旁组织,在胆管癌细胞系 TFK-1、RBE、HuH28、HuCCT-1、QBC939及正常胆管上皮细胞 HIBEpic中检测 miR-28-5p的表达水平。采用 Kaplan-Meier曲线和 Cox回归分析 miR-28-5p与患者预后的关系。采用 CCK-8和 Transwell实验分析 miR-28-5p对细胞增殖、迁移和侵袭的影响。结果:胆管细胞癌组织和细胞系中 miR-28-5p表达水平均显著高于癌旁组织和正常上皮细胞。miR-28-5p低表达与患者的不良预后显著相关。下调 miR-28-5p可促进胆管癌细胞的增殖、迁移和侵袭,而过表达 miR-28-5p则可抑制细胞的增殖、迁移和侵袭。结论:miR-28-5p有作为胆管细胞癌患者的预后标志物和潜在治疗靶点的前景。

Abstract:
Objective To investigate the relationship between miR-28-5p expression and the prognosis of cholangiocarcinoma patients, as well as its functional role in tumor occurrence and development. Methods A total of 112 cholangiocarcinoma patients who underwent surgical resection were included in this study. The expression levels of miR-28-5p were detected in 112 pairs of cholangiocarcinoma tissues and adjacent non-cancerous tissues, as well as in CCA cell lines (TFK-1, RBE, HuH28, HuCCT-1, QBC939) and normal biliary epithelial cells (HIBEpic). The relationship between miR-28-5p and patient prognosis was analyzed using Kaplan-Meier curves and Cox regression analysis. The effects of miR-28-5p on cell proliferation, migration, and invasion were assessed using CCK-8 and Transwell assays. Results The expression levels of miR-28-5p in cholangiocarcinoma tissues and cell lines were significantly higher than those in adjacent non-cancerous tissues and normal epithelial cells. Additionally, low expression of miR-28-5p was significantly associated with poor prognosis in patients. Downregulation of miR-28-5p promoted proliferation, migration, and invasion of cholangiocarcinoma cells, while overexpression of miR-28-5p inhibited these processes. Conclusion miR-28-5p holds promise as a prognostic marker and potential therapeutic target for cholangiocarcinoma patients.
