清胰颗粒对轻症急性胰腺炎大鼠炎症损伤及核因子-κB 信号通路的影响

摘要:
目的探讨清胰颗粒对轻症急性胰腺炎(MAP)大鼠炎症损伤的修复作用及对核因子(NF)-κB信号通路的影响。方法:选取Wistar大鼠50只,按随机数字法分为对照组、模型组和治疗组,模型组和治疗组再分为2个亚组,分别在建模后12 h和24 h取材,每组各10只。采用注射雨蛙素法建立大鼠MAP模型,治疗组给予清胰颗粒灌胃,模型组给予等量生理盐水灌胃。观察胰腺组织病理变化,检测大鼠血清淀粉酶(AMY)、脂肪酶(LIP)活力及Ca2+浓度,采用ELISA法检测血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1、IL-6水平;采用Western blot法检测胰腺组织中NF-κB蛋白的表达水平。结果:胰腺组织病理评分模型组高于对照组,同时点病理评分治疗组均低于模型组(P<0.05)。模型组血清AMY、LIP活力,炎症因子IL-1β、IL-6、TNF-α浓度,NF-κB蛋白活化水平均高于对照组(P<0.05),治疗组血清AMY、LIP活力,IL-1β、IL-6、TNF-α浓度,NF-κB蛋白活化水平低于模型组(P<0.05);模型组血清Ca2+浓度低于对照组,治疗组血清Ca2+浓度高于模型组(P<0.05)。结论:清胰颗粒可以下调MAP大鼠胰腺组织的NF-κB信号通路的表达,减少炎症因子IL-1β、IL-6、TNF-α的释放,减轻胰腺组织损伤,并促进胰腺组织修复。

Effects of Qingyi granules on inflammatory injury and NF-κB signaling pathway in mild acute pancreatitis rats

Abstract:
Objective To explore the repair effect of Qingyi granules on inflammatory damage and the influence of NF-κB signaling pathway in rats with mild acute pancreatitis (MAP). Methods 50 Wistar rats were selected and divided into groups according to the random number method, including the control group, the model group and the treatment group. The model group and the treatment group was further divided into two subgroups, with sample collections at 12 and 24 hours post-modeling,with 10 rats in each group. A rat model of MAP was established by injecting cerulein. The treatment group was given Qingyi granules by gavage, and the model group was given an equal amount of normal saline by gavage. Pancreatic tissue changes were meticulously observed under a microscope. Serum amylase, lipase, and calcium levels were detected; TNF-α, IL-1β, IL-6 levels were measured using ELISA, and protein expression levels were assessed using Western Blot. Results The pancreatic tissue pathology score in the model group was higher than that in the control group, while the treatment group was lower than the model group (P<0.05). The serum AMY and LIP activities, inflammatory factor IL-1β, IL-6, and TNF-α concentrations, and NF-κB protein activation level in the model group were all higher than those in the control group (P<0.05). The serum AMY and LIP activities, the concentrations of IL-1β, IL-6, TNF-α, and NF-κB protein activation level were lower than those of the model group (P<0.05); the serum Ca2+ concentration of the model group was lower than that of the control group, and the serum Ca2+ concentration of the treatment group was higher than that of the model group (P<0.05). Conclusion Qingyi granules can down-regulate the expression of NF-κB signaling pathway in pancreatic tissue of MAP rats, reduce the release of inflammatory factors IL-1β, IL-6, and TNF-α, reduce pancreatic tissue damage, and promote pancreatic tissue repair.
