Effect and mechanism of Astragalus membranaceus combined with Ligustrazine on reducing volume loss of free dermal fat flap

Abstract:
Objective To investigate the effect and mechanism of astragalus membranaceus combined with ligustrazine on reducing the volume loss of free dermal fat flap, and to explore the best adjuvant medication. Methods The free dermal fat flap from the abdomen of female SD rats was transplanted into the chest and successfully modeled. Then, the rats were divided into four groups (20 in each group): control group, daily intraperitoneal injection of normal saline; Astragalus membranaceus injection (AM) group was intraperitoneally injected with Astragalus injection (2 g/mL) every day; Ligustrazine injection group, daily intraperitoneal injection of ligustrazine injection (2.4 mg/mL); AM + ligustrazine group was intraperitoneally injected with a mixture of AM injection and ligustrazine injection 1:1 every day for four weeks. The grafts were removed immediately, 1 week, 2 weeks and 4 weeks after surgery. The weight and thickness of the dermal fat flap and the degree of oxidative damage were measured at each time point. The expressions of vascular endothelial cell growth factor (VEGF) and highly glycosylated type I transmembrane glycoprotein (CD34) were measured by immunohistochemistry and western blotting. Results All dosage groups can increase the weight of the free leather fat flap and thickness (P?0.001). The activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) was increased, and the content of malondialdehyde (MDA) in dermal fat was decreased (P<0.05). All dosage groups were increased significantly in 4 weeks for the free leather fat flap of tissue VEGF expression (P?0.05), including AM injection combined with ligustrazine injection significantly increased the expression of CD34 (P=0.002). Conclusion Our research provides a novel insight into the effects of AM and ligustrazine injection on delaying the atrophy of free dermal fat.
