黄芪在抗菌肽Brevinin-2Ta治疗创面感染耐药鲍曼不动杆菌中的增效机制研究

摘要:
目的 探讨中药黄芪在抗菌肽Brevinin-2Ta治疗创面感染耐药鲍曼不动杆菌中的增效作用及机制。方法:选取SD大鼠60只,采用皮肤切开注菌法建立皮肤感染泛耐药鲍曼不动杆菌溃疡模型,将模型大鼠随机分为3组:空白组、抗菌肽组及抗菌肽+黄芪组,分别于造模成功后第3天、第15天取大鼠伤口观察愈合情况,取伤口处组织,在电镜下观察细菌形态,并用荧光PCR法测定carO基因表达水平。结果:用药后第3天,空白组的carO相对定量为28.770±8.829;抗菌肽组carO相对定量为14.721±3.879;抗菌肽+黄芪组carO相对定量为11.768±2.752。抗菌肽组、抗菌肽+黄芪组carO表达较空白组明显受抑制(P<0.05),而抗菌肽组与抗菌肽+黄芪组carO表达无明显差异(P>0.05)。用药后第15天,空白组carO相对定量为28.113±4.996;抗菌肽组carO相对定量为17.490±2.648;抗菌肽+中药组carO相对定量为11.082±2.476,空白组、抗菌肽组及抗菌肽+黄芪组间差异均有统计学意义(P<0.05)。用药后电镜下显示:第3天时抗菌肽组菌体细胞膜结构被破坏,相互粘连,抗菌肽+黄芪组可见大量细菌坏死粘连;第15天时抗菌肽组及抗菌肽+黄芪组可见细菌大量坏死,抗菌肽+黄芪组优于抗菌肽组。结论:中药黄芪在抗菌肽Brevinin-2Ta治疗创面感染耐药鲍曼不动杆菌中具有一定的增效作用,其机制可能与carO表达受抑制有关。

Study on synergistic mechanism of astragalus in Brevinin-2Ta for wound infection-resistant Acinetobacter baumannii

Abstract:
Objective To investigate the synergistic effect and mechanism of the traditional Chinese medicine Astragalus astragalus in the treatment of antibacterial peptide Brevinin-2Ta in the treatment of wound infection resistant Acinetobacter baumannii. Methods 60 SD rats were randomly divided into blank group, antibacterial peptide group and antimicrobial peptide + astragalus group, and the wound wound of rats was taken on the 3rd and 15th days after successful modeling, the wound of the wound was taken, the tissue at the wound was taken, the bacterial morphology was observed under electron microscopy, and the expression content of carO gene was determined by fluorescent PCR. Results The carO of the blank group on day 3 after administration was 28.770±8.829, antimicrobial peptide group was 14.721±3.879; antimicrobial peptide + astragalus group was 11.768±2.752. The expression of carO in the antimicrobial peptide group and the antimicrobial peptide + astragalus group was significantly inhibited compared with the blank group (P<0.05), while there was no significant difference in the expression of carO between the antimicrobial peptide group and the antimicrobial peptide + astragalus group (P>0.05). At the 15th day after administration, the carO of the blank group was 28.113±4.996; antimicrobial peptide group was 17.490±2.648; antimicrobial peptide + traditional Chinese medicine group was 11.082±2.476, and there were statistical differences between the blank group, antimicrobial peptide group and antimicrobial peptide + astragalus group (P<0.05). After treatment, electron microscopy showed that on the 3rd day, the bacterial cell membrane structure of the antibacterial peptide group was destroyed and adhered to each other, and a large number of bacterial necrotic adhesions could be seen in the antibacterial peptide + astragalus group; On the 15th day, a large amount of bacterial necrosis was seen in the antibacterial peptide group and the antibacterial peptide + astragalus group, and the antibacterial peptide + astragalus group was better than the antibacterial peptide group. Conclusion The traditional Chinese medicine Astragalus has a certain synergistic effect in the treatment of antibacterial peptide Brevinin-2Ta in the treatment of wound infection resistant Acinetobacter baumannii, and its mechanism may be related to the inhibition of carO expression.
