Mechanisms of isorhynchophylline on proliferation and migration of non-small cell lung cancer A549 cells

Abstract:
Objective To investigate the effects of isorhynchophylline on the growth, motility, and ferroptosis of non-small cell lung cancer (NSCLC) cells, as well as its influence on the Wnt/β-catenin signaling pathway. Methods The human NSCLC cell line A549 was selected for grouped experiments. A549 cells were treated with 0 and 2 mg/L concentrations of isorhynchophylline for 24 hours. Cell growth was evaluated using CCK-8 and clonogenic assays, while cell motility was assessed through wound healing assays. Ferroptosis was measured using immunoblotting and iron detection kits, and the activation of the Wnt/β-catenin signaling pathway was evaluated by Western blotting. Results Isorhynchophylline significantly inhibited the growth and motility of human NSCLC cells. Additionally, isorhynchophylline promoted ferroptosis in NSCLC cells. The antitumor effects of isorhynchophylline were associated with the inhibition of the Wnt/β-catenin signaling pathway, which is crucial for cell survival and growth. Conclusion: Isorhynchophylline exerts antitumor effects on NSCLC cells by inhibiting cell growth and motility, promoting ferroptosis, and suppressing the Wnt/β-catenin signaling pathway.
