Research of the protective effect of Xuebijing on the liver of septic thrombocytopenic rats

Abstract:
Objective To investigate the effect of Xuebijing on the liver injury in the septic thrombocytopenic rats. Methods Rats were randomly divided into four groups: control group (Sham), cecum ligation perforation model group (CLP), Xuebijing treatment group (XBJ) and Xuebijing prevent intervention group (pre-XBJ). The rats of CLP group,XBJ group and pre-XBJ group were performed cecal ligation and perforation to induce sepsis thrombocytopenia model and the Sham group rats were only opened without ligation and perforation. Before the operation, the pre-XBJ group rats were giving Xuebijing by tail vein injection and the other groups were giving equal volume of normal saline. The XBJ group rats were giving Xuebijing by tail vein injection at 0 h and 12 h after operation, and the other groups were given an equal volume of normal saline.Blood was collected from the abdominal aorta at the 24 h after operation to detect platelet counts, and ELISA was used to detect liver function related indicators, TNF-aand other inflammatory factors. HE staining was used to observe liver injury and immunohistochemistry to detect platelet retention in the liver. Results The platelet count of rats in the CLP group was significantly lower than that in the Sham group, while the platelet count of rats in the XBJ intervention groups increased than the CLP group (P<0.05). The AST,ALT and TB in the XBJ intervention groups were all lower than the CLP group (P<0.05), and the expression level of TNF-a, IL-6 and IL-18 were also reduced in the XBJ group and pre-XBJ group than that in the CLP group(P<0.05). The pulmonary tissue edema, inflammatory cell infiltration and platelet retention in the liver in the XBJ group and pre-XBJ group were all reduced compared with the CLP group. Conclusion Xuebijing can relieve the reduction of platelet count in septic thrombocytopenic rats, improve liver damage and platelet aggregation and adhesion in the liver, and has a certain protective effect on the liver of septic thrombocytopenic rats.
