Mechanism of compound seabuckthorn seed oil accelerating postoperative wound repair of perianal abscess in rats

Abstract:
Objective To study the effect of compound seabuckthorn seed oil on accelerating wound healing after perianal abscess surgery in rats. Methods A total of 50 SD model mice were used to successfully prepare postoperative wounds of perianal abscess. They were randomly divided into five groups: G1 model group (given distilled water); G2 positive control group (given potassium permanganate); G3 treatment group (low dose group of compound seabuckthorn seed oil nano-emulsion); G4 treatment group (middle dose group of compound seabuckthorn seed oil nano-emulsion); G5 (high dose group of compound seabuckthorn seed oil nano-emulsion). After modeling, the wounds of experimental rats in each group were evenly smeared with corresponding experimental drugs twice a day for 14 days. Observe the wound healing of rats in each group after 5 d, 8 d and 14 d of treatment, calculate the wound healing rate of rats in each group and take the wound tissue of rats in each group respectively, and analyze the expression of EGF, phosphorylated ERK1/2, IL-8 and PEG2 in the wound with immunohistochemical S-P method. Results The wound healing rate of rats in G5 group, G4 group, G3 group, G2 group and G1 group decreased in turn at 5 d, 8 d and 14d after wound treatment (all P<0.05). 14 days after surgery, IL-8 expression in rat wounds G2G4>G3>G2>G1 (all P<0.05). Conclusion Compound seabuckthorn seed oil nano-emulsion can effectively promote wound healing after perianal abscess surgery in rats, which may be related to the inflammatory mediators of IL-8 and PEG2, and its activation of "EGF-ERK" pathway can promote the growth, proliferation and differentiation of wound keratinocytes and fibroblasts.
