Investigation of the mechanism of Jichuan decoction intervention on serum VIP and colonic C-Kit to improve slow-transmission constipation in rats

Abstract:
Objective To investigate the effect and mechanism of Jichuan decoction on rat models of slow-transit constipation. Methods Forty SD rats were randomly divided into normal group, model group, Jichuan fried high, medium and low dose group, with 8 rats in each group. The normal group was given 10 mL/(kg·d)normal saline gavage for 14 days; The remaining groups were gavaged with compound dipheNOxylate [10 mL/(kg·d)] at a concentration of 1.5 mg/mL for 14 days to establish an STC model. Jichuan decoc tion gavage of 1.85 g/kg, 3.7 g/kg and 7.4 g/kg was given to the low, medium and high dose groups of Jichuan decoction for 14 days, and the normal group and the model group were gavaged with the same volume of distilled water. The time of first melena and the length of carbon powder propagation in the colon were determined in each group. The ELISA method measures the content of C-Kit and serum vasoactive intestinal polypeptide(VIP) in colonic tissue. Results Compared with the normal group, the first melena time increased, the concentration of VIP in serum increased significantly, the length of carbon end propeller and the content of C-Kit protein in the colon decreased (all P<0.05). Compared with the model group, the first melena time and serum VIP concentration of rats in the low, medium and high dose groups in Jichuan fried significantly decreased, and the length of intestinal carbon powder advance and colonic C-Kit protein content increased significantly (all P<0.05), and compared with the Jichuan fried group, the first melena time and serum VIP concentration in the high-dose group were significantly reduced, the length of intestinal carbon powder advancement and colonic C-Kit content were significantly increased, and the difference between the medium and low dose groups was significantly higher (P<0.05). Conclusion Jichuan decoction can promote intestinal motility and improve constipation symptoms, and its mechanism of action may be related to increasing the expression of C-Kit in colon tissues and reducing the content of VIP in serum.
