Study on expression of high mobility group protein B1 and DJ-1 in bladder cancer and clinical related problems

Abstract:
Objective To investigate the expression and clinical significance of high mobility group protein B1(HMGB-1) and Parkinson's disease associated protein (DJ-1) in different tissues of bladder cancer. Methods From January 2005 to January 2015, 58 cases of bladder cancer treated in the Second Hospital of Tianjin Medical University and Tianjin Nankai Hospital were collected as the experimental group, and their adjacent normal tissues as the control group. Immunohistochemical method was used to detect the expression rates of HMGB-1 and DJ-1 proteins in bladder cancer tissues and adjacent tissues. The relationship between HMGB-1, DJ-1 and the clinicopathological features of bladder cancer and the survival of patients was analyzed. Spearman correlation analysis was used to analyze the correlation between HMGB-1 and DJ-1 expression in bladder cancer, and Cox multivariate analysis was used to analyze the prognostic factors of bladder cancer patients. Results The positive expression rates of HMGB-1 and DJ-1 in bladder cancer were higher than those in adjacent tissues, and the difference was statistically significant (P<0.05). The expression level of HMB-1 protein is related to pathological classification, histological differentiation, lymph node metastasis and TNM stage, and the worse the tumor differentiation, lymph node metastasis and stage, the higher the HMB-1 expression (P<0.05). The expression of DJ-1 was related to the number of tumors, lymph node metastasis and TNM stage. The expression level of DJ-1 protein in multiple tumors, high TNM stage and lymph node metastasis group was significantly higher than that in single tumor, low stage and no local metastasis group (P<0.05). Spearman correlation analysis showed that the expression levels of HMGB-1 and DJ-1 were positively correlated (r=0.421, P<0.05). Patients with high expression of HMB-1 and DJ-1 protein had a shorter disease-free survival time than those with low expression of HMB-1 and DJ-1 protein (P<0.05). Cox regression model analysis showed that HMGB-1, DJ-1, adenocarcinoma, squamous cell carcinoma, urothelial carcinoma, histological differentiation, lymph node metastasis and TNM stage can be used as independent risk factors to predict the prognosis of bladder cancer. Conclusion The detection of HMGB-1 and DJ-1 expression levels in bladder cancer is helpful to predict and evaluate the invasion, metastasis and prognosis risk of bladder cancer, and provides an important basis for the rational choice of perioperative treatment for bladder cancer.
