Correlation of Serum miR-615-5p and SHH Expression with Clinicopathological Features and Prognosis in Patients with Colorectal Cancer

Abstract:
Objective To investigate the relationship between the serum microRNA (miR)-615-5p and sonic hedgehog (SHH) expression and the clinicopathological characteristics and prognosis of patients with colorectal cancer (CRC). Methods A total of 136 CRC patients in our hospital were gathered as the CRC group, and the healthy persons (100 cases) who underwent physical examination were regarded as the healthy group. The differences in the expression levels of serum miR-615-5p and SHH between the two groups, and the relationship between the different expression levels of miR-615-5p and SHH and the clinicopathological characteristics of the patients were analyzed, and the related factors affecting the prognosis of CRC were analyzed; the correlation between miR-615-5p and SHH was analyzed by Pearson method; and the relationship between serum miR-615-5p, SHH expression and prognosis of CRC patients was analyzed by Kaplan-Meier survival curve. Results The expression level of serum miR-615-5p in the CRC group (0.57±0.13) was lower than that in the healthy group (1.02±0.05), and the expression level of SHH (1.85±0.31) was higher than that in the healthy group (0.98±0.10) (P < 0.05); miR-615-5p was negatively correlated with SHH (r=-0.631, P < 0.05). The expression levels of serum miR-615-5p and SHH were correlated with tumor T stage and N stage (P < 0.05). T stage and SHH were independent risk factors for CRC prognosis, and miR-615-5p was a protective factor for CRC prognosis. Survival analysis showed that the cumulative survival rate of the miR-615-5p high expression group was higher than that of the miR-615-5p low expression group, and the cumulative survival rate of the SHH low expression group was higher than that of the SHH high expression group (P < 0.05). Conclusion The expression of miR-615-5p in serum of CRC patients was decreased, while the expression of SHH was increased, both of which were related to T stage and N stage shift. CRC patients with low expression of miR-615-5p or high expression of SHH had poor prognosis
