Protective Effect of Total Glucosides of Paeony on Mitochondria of Interstitial Cells of Cajal in Rats with Intestinal Ischemia-reperfusion

Abstract:
Objective To explore the protective effect of total glucosides of paeony on the mitochondria of interstitial cells of Cajal in intestinal ischemia-reperfusion (IIRI) rats. Methods Forty-five rats were randomly divided into 3 groups (sham operation group, model group and total glucosides of paeony group). Seven days before model establishment, the total glucosides of paeony were given by gavage of total glucosides of paeony.The sham operation group and the model group were given the same dose of normal saline by gavage, 1 time/day.Results Compared with the sham operation group, the small intestine propulsion rate of the model group was significantly reduced at 12 and 24 hours after the operation (P <0.01). Compared with the model group, the small intestinal propulsion rate of rats in the total glucosides of paeony group increased significantly at 12 and 24 hours after surgery (P <0.01). Under the optical microscope, the intestinal damage of the rats in the total glycosides of paeony group significantly reduced compared with the model group. The intermucosal congestion, edema and congestion significantly improved. Under the lens, the interstitial cells of Cajal in the total glycosides of paeony group significantly improved. The mitochondrial swelling reduced, autophagosomes reduced and gap junctions were visible. Compared with the sham operation group, the expression levels of SCF, C-kit and Cx43 protein in the small intestine of the model group significantly reduced. Compared with the model group, the SCF, C-kit and Cx43 protein expression levels of rats in the total glucosides of paeony group increased signi.cantly (P<0.01). Conclusion The pretreatment of total glucosides of paeony which improved the intestinal injury were induced by IIRI. It had a protective effect on the mitochondrial structure and functional stability of interstitial cells of Cajal, which may be related to the C-kit/SCF signal pathway.
