Effect of Yiqi Huayu Decoction on Nerve Repair in Rats with Diabetic Ulcer

Abstract:
Objective To investigate the effect of Yiqi Huayu Decoction in regulating PI3K/AKT/mTOR signal pathway on peripheral nerve repair in rats with diabetic ulcer. Methods Thirty healthy male (SPF grade) SD rats were established a diabetic peripheral neuropathy ulcer model, then they were randomly divided into model group, Yiqi Huayu Decoction group, Yiqi Huayu Decoction+PI3K inhibitor group, Yiqi Huayu Decoction+mTOR inhibitor group. The sciatic nerve morphology of rats on the 21st day after administration was observed and the nerve conduction velocity, wound neuropeptide P substance (SP), NGF protein and NGF mRNA content were detected. Results Compared with the control group, the sciatic neuropathy of the model group was more severe. The nerve conduction speed was slowed down, and the contents of SP, NGF protein and NGF mRNA were decreased, which the difference were statistically significant (P<0.05). Compared with the model group, the sciatic neuropathy of the Yiqi Huayu Decoction group reduced, nerve conduction speed increased, SP, NGF protein and NGF mRNA content increased (P<0.05). Compared with Yiqi Huayu Decoction group, the degree of sciatic neuropathy in the Yiqi Huayu Decoction+PI3K inhibitor group and Yiqi Huayu Decoction+mTOR inhibition group was more serious, nerve conduction speed was slowed down, and the contents of SP, NGF protein and NGF mRNA were decreased, which the difference were statistically significant (P<0.05). Conclusion Yiqi Huayu Decoction may improve the expression of SP and NFG by regulating the PI3K/AKT/ mTOR signal pathway, and promote the repair of peripheral nerves in diabetic ulcer rats.
