Effect of Zihuang Shengji Ointment on Angiogenesis and Expression of VEGF and Ang-1 in Chronic Infection Wound

Abstract:
Objective To investigate the molecular mechanism of Zihuang Shengji Ointment in promoting angiogenesis of chronic infectious wounds. Methods 84 SD rats were randomly divided into 4 groups: blank group, model group, rb-bFGF group, Zihuang Shengji Ointment group. After successfully establishing chronic infectious wound models, all groups were treated with 1 mL of fecal fluid on the wound surface and kept for half an hour before clean dressing everyday, in addition to blank group. The effect of wound healing was observed at 3, 7 and 14 days after treatment, and the number of capillaries was observed too. The expressions of VEGF and Ang-1 were detected by ELISA. Results Zihuang Shengji Ointment had less blood and fluid in the wound, and the granulation tissue grew well. On the 7th and 14th day, a large number of new blood vessels were found in the wound, and the wound healing was significantly faster than that in the model group and rb-bFGF group (P<0.01). On the 7th and 14th day, compared with the model group and rb-bFGF group, VEGF and Ang-1 in Zihuang Shengji Ointment group were significantly increased (P<0.01). Conclusion Zihuang Shengji Ointment can significantly promote wound healing, shorten wound healing time and promote angiogenesis in rats with chronic infectious wounds. Its mechanism may be related to the rapid and effective up-regulation of VEGF and Ang-1.
