Effect of Serum Containing Taohe Chengqi Decoction on LPS Induced Proliferation of Aortic Endothelial Cells and Expression of Related Factors

Abstract:
Objective To investigate the effect of serum containing Taohe Chengqi Decoction on the proliferation of aortic endothelial cells induced by lipopolysaccharide (LPS) and the expression of related factors. Methods 30 male SD rats were randomly divided into two groups: normal group and medicated serum group with 15 rats in each group. Medicated serum group was given Taohe Chengqi Decoction by gavage, and normal group was given the same dose of normal saline by gavage. After 7 days, blood was taken from abdominal aorta and serum was centrifuged for separate use. CCK-8 method and flow cytometry were used to detect the effects of different concentrations of drug containing serum on the proliferation and apoptosis of endothelial cells. The optimal intervention concentration was 10 μmol/L. The cells were divided into blank control group (10% blank serum), blank serum group (10% medicated serum), model control group (LPS+10% blank serum) and model serum group (LPS+10% medicated serum). Western-blotting was used to detect the expression of phosphatidylinositol-3 kinase (PI3K), protein kinase B (Akt), transforming growth factor-β (TGF-β) and pseudo protein kinase 3 (TRIB3) in aortic endothelial cells after intervention. Results Compared with blank serum group and blank control group, the expression of PI 3 K, Akt and TGF-β in model control group decreased (P< 0 . 05 ) and the expression of TRIB 3 increased (P<0.05). After cultured in Taohe Chengqi Decoction drug-containing serum, compared with model control group, the expression of PI3K, AKT,TGF-β in model serum group increased and the expression of TRIB3 decreased (P<0.05). Conclusion The serum containing Taohe Chengqi Decoction can inhibit the expression of TRIB3 and promote the expression of PI3K, Akt and TGF-β. The serum containing Taohe Chengqi Decoction can promote the regeneration of injured endothelial cells, which may be related to PI3K / Akt signaling pathway.
