Effect of Huoxuo Tongjiang Prescription on NF-κB/MAPK Signal Pathway of Esophageal Mucosa in Rats with RE

Abstract:
Objective The aim of this study was to investigate the effect of Huoxue Tongjiang prescription on the NF-κB/MAPK signal pathway of esophageal mucosa in rats model of RE, and explore the mechanism of traditional Chinese medicine in the treatment of RE furtherly. Methods 40 rats were randomly divided into 4 groups: control group, model group, western medicine group and Chinese medicine group. The control group was fed without treatment; the other 3 groups were treated with RE (the method was modified partial cardiotomy and external partial pylorus ligation). After operation, the model group was given 0.9% normal saline for 14 days, and the western medicine group was given western medicine suspension (domperidone+ omeprazole+ aluminum magnesium carbonate), and the Chinese medicine group was given Huoxue Tongjiang prescription, all for 14 days. The general conditions of the rats were observed. On the 7th and 14th day, half of the rats in each group were killed. The pathological score, p38MAPK and NF-κB p65 immunohistochemical staining were observed and the serum levels of IL-6 and TNF-α were measured. Results On the 7th day of treatment, the scores of esophageal mucosa in western medicine group and Chinese medicine decreased significantly compared with those of the model group (P<0.05), indicating that western medicine and Chinese medicine have curative effects on RE model rats, but there is nostatistical difference between western medicine group and traditional Chinese medicine (P > 0.05).On the 14th day of treatment, the general specimens of esophageal mucosa in western medicine group and traditional Chinese Medicine decreased. Compared with the model group, the difference was more signi?cant (P < 0.01). Compared with the control group, there was no statistical difference (P > 0.05). There was no statistical difference between the western medicine group and Chinese medicine group (P > 0.05). Compared with the control group, the expression of p38MAPK and NK-κB inflammatory pathway, IL-6 and TNF - α in the serum increased signi?cantly (P < 0.01). On the 7th day of treatment, the expression of the above-mentioned indexes decreased significantly in the western medicine group and the Chinese medicine, which compared with the model group (P < 0.05). On the 14th day of treatment, there was no significant differencethe expression of the above-mentioned indexes between the western medicine group and the Chinese medicine group(P > 0.05). Conclusion Huoxue Tongjiang prescriptiontreatment can effectively improve the inflammation and histopathological changes of esophageal mucosa, inhibit the p38MAPK / NF-κB inflammatory signal pathway, reduce the production of inflammatory factors and treat RE by controlling the immune inflammation of RE.The effect is positively correlated with treatment time.
