Study on the Mechanism of Anal Pruritus Lotion in the Treatment of Perianal Eczema based on Network Pharmacology

Abstract:
Objective Preliminary study on the mechanism of anal pruritus lotion in the treatment of perianal eczema based on network pharmacology. Methods The TCMSP database was used. After screening the component information according to DL ≥ 0.18 and ALogP<0.5, searching for drug-related targets, a total of 258 drug targets were obtained. Searching for genes related to “eczema” disease in the GeneCards database, a total of 2 736 related genes were obtained. There are 114 targets for drugs and diseases. The protein protein interaction (PPI) relationship was obtained by string database and visualized by Cytoscape software. According to the “degree”value of protein interaction, the key targets were obtained, which suggested that the anal pruritus lotion may participate in cytokine-mediated cell signal transduction to initiate the effect. The “Bioconductor”installation package in R software was used to analyze the GO and KEGG pathways of the target. Results Network analysis results showed that the anal pruritus lotion mainly induced immune activation through cytokines and receptors, and might exert its biological activity through signal pathways such as IL-17, TNF, Toll receptor, JAKSTAT, Th1 and Th2 cell differentiation. Conclusion This study comprehensively applied the method of network pharmacology to predict the mechanism and molecular targets of anal pruritus lotion in the treatment of eczema, which is the basis for further experimental research and identification.
