Effect of Weinongzhangrou Method on Expression of VEGF, Notch1 and TGFβ1 in Granulation Tissue of Rats with Chronic Ulcer and its Histopathology

Abstract:
Objective To observe the effects of Weinongzhangrou method on the expression of VEGF, Notch1 and TGF-β1 in the granulation tissue of rats with chronic ulcer. Methods Totally 24 rats were randomly divided into four groups: normal group, model group, control group and treatment group. The rats in normal group had no wound; The other three groups of rats used the compound factors superposition method of “hormone intervention-skin defect-bacterial infection” to duplicate the model of chronic skin ulcer. The rats in model group were given normal saline treatment, the rats in control group were given vaseline gauze external treatment, and the rats in treatment group were given external application of Shengji Xiangpi Plaster, once daily for 14 d. After 3, 7 and 14 d of treatment, the rat wound granulation tissues of rats were taken and stained by HE to observe the histopathological changes, and the expression of VEGF, TGF-β1 and Notch 1 were detected by immunohistochemistry. Results After treatment of 14 d, the wound healing in the treatment group was basically healed (0.012±0.004) cm2, and the difference was statistically signi.cant (P<0.05 or 0.01) compared with the model group and control group. In early and medium terms of wound healing (3 to 7 d), the expression levels of TGF-β1, VEGF and Notch 1 in the wound tissue of the treatment group increased compared with the model group and control group, but the difference was statistically significant (P<0.05); in the late stage of wound healing (14 d), there was no significant difference in each index (P>0.05) Conclusion Weinongzhangrou method may be used to regulate the expression of VEGF, Notch 1 and TGF-β1 in wound surface, which can promote the growth of granulation and wound healing.
