Optimized Compatibility Ratio of Radix Astragali and Radix Paeoniae Rubra based on Pharmacodynamics Study

Abstract:
Objective To explore the optimal compatibility ratio of Radix Astragali and Radix Paeoniae Rubra in Danqi Hemiplegia Capsule for treating the acute stage of ischemic cerebrovascular disease. Methods Four different compatible proportions of Radix Astragali and Radix Paeoniae Rubra were chosen, with carotid thrombosis time as testing index, a valid sample was selected. Then healthy Wistar rats were selected for the preparation of hyperviscosity rat model, and the dosages of 2 . 5 , 5 and 10 mL/kg were given respectively as the low-, mid- and high-dose groups, to observe the effects of different dosages of Astragalus membranaceus and Radix Paeoniae Rubra on the blood viscosity and plasma fibrinogen content of hyperviscosidemia rats. Results When the compatibility ratio of Radix Astragali and Radix Paeoniae Rubra was 1:3, the time of carotid artery thrombosis could be signi?cantly prolonged. The effects of different doses of the concentrate on the blood viscosity were found that the mid- and high-dose concentrate signi?cantly decreased the whole blood viscosity at different shear rates, and the low-, mid- and high-dose concentrate significantly reduced the hematocrit to ( 48 . 1 ± 4. 3)%, ( 47. 5 ± 3 . 4 )%, and ( 42 . 2 ± 2 . 4 )%. As the dose increased, the effect was enhanced. High dose group could signi?cantly reduce plasma ?brinogen content to (1.3±0.3) g/L.Conclusion The compatibility ratio of Radix Astragali and Radix Paeoniae Rubra is 1:3, and the dosage of 10 mL/kg is obviously effective in promoting blood circulation.
